Preparation of stains-
Working MG solution:-
Prepare MG (May-Grandwal) stain with D/W in ratio of 1:1(
take same volume of stain and D/W)
Working Giemsa solution:-
Giemsa stain and Distilled water should be taken in ratio of
1:10(1ml Giemsa stain + 9ml D/W)
Procedure:-
1. Air dried slides are fixed in alcohol.
2. Take out the slides and air dried .
3. The smear is covered with working MG stain for 10 minutes.
4. Discard the stain after 10minutes.
5. Cover the smear with working Giemsa stain keep for
15minutes
6. Wash the slide in running tape water and allowed to air
dry.
7. Dried slides are dipped in xylene and mounted in DPX.
Nucleus:- blue
Cytoplasm and other components:- Pale/ light blue
