This method is used for samples-
such as vaginal, ectocervical , endocervical, bronchial wash etc.
Fixative used are- isopropanolol/isopropyl alcohol/95% alcohol.
This method of staining also called as Rapid PAP method.
Procedure:-
1. Wet smears prepared on the clean glass slide are dipped in couplin jar which contain fixative.
2. The smears are taken out of the jar and allowed to dry.
3. Wash with tape water.
4. Dipped in cytoplasmic stain for 2-3 minutes.
[cytoplasmic working stain-2A(OG-6)25 ml + 2B(light green SF-yellow)25 ml ]
5. Wash in tape water.
6. Blot out excess water and dip in 95%alcohol for dehydration.
7. Blot and dry.
8. Dip in xylene before mounting with coverslip and DPX.
Result:-
Nucleus:- blue
Cytoplasm:-varying shades of pink , blue, yellow green - grey.
